RETATRUTIDE

Retatrutide is an investigational, once-weekly injectable medication developed by Eli Lilly and Company, representing a next-generation advancement in the treatment of obesity and related metabolic conditions. As a first-in-class triple hormone receptor agonist, retatrutide simultaneously activates three key receptors:

• GLP-1 (glucagon-like peptide-1) receptor — helps regulate appetite, slow gastric emptying, and improve blood sugar control
• GIP (glucose-dependent insulinotropic polypeptide) receptor — enhances insulin secretion and supports metabolic balance
• Glucagon (GCG) receptor — promotes increased energy expenditure and fat metabolism

This unique triple-action mechanism sets retatrutide apart from dual-agonist therapies, enabling synergistic effects on appetite suppression, calorie burning, glycemic control, and overall body composition.

In clinical trials, including Phase 2 and the first successful Phase 3 study (TRIUMPH-4), retatrutide has demonstrated substantial results when used alongside a healthy diet and physical activity:

• Up to 24.2% mean body weight reduction over 48 weeks in Phase 2
• Up to 28.7% average weight loss (equivalent to about 71.2 lbs in some participants) over 68 weeks in Phase 3 among adults with obesity or overweight
• Additional benefits such as reduced liver fat, improved lipid profiles (e.g., lower triglycerides and non-HDL cholesterol), decreased blood pressure, and — in specific populations — significant relief from knee osteoarthritis pain and improved physical function

In trials, Retatrutide is administered as a subcutaneous injection once per week, with a half-life of approximately 6 days supporting convenient dosing. Common side effects in trials have been primarily gastrointestinal (e.g., nausea, diarrhea), dose-related, and generally mild to moderate, often mitigated with gradual dose escalation.

Currently, retatrutide is in ongoing Phase 3 clinical trials (including the TRIUMPH program). Additional data readouts are expected throughout 2026, with potential future availability pending successful completion of studies and regulatory review for indications including obesity, type 2 diabetes, and weight-related complications.
